ABSTRACT Gingival recession is the apical displacement of the gingival margin in relation to the cement-enamel junction, leading to root exposure. This condition may cause cervical dentin hypersensitivity (CDH), compromising the esthetics and interfering with quality of life. The objective of this study was to evaluate the effect of root coverage on the CDH, aesthetics and quality of life of patients with gingival recessions class I and II Miller. The secondary aim was to investigate the etiologic factors of gingival recessions. Twenty-two patients aged 18-50 years presented 25 gingival recessions in sensitive upper canines and premolars. The following parameters were assessed: width and height of the gingival recessions were assessed with a compass needle point and measured with a caliper. It was assessed the height and thickness of keratinized gingiva, plaque index, bleeding on probing, probing depth and clinical attachment level. The CDH was assessed by evaporative (air jet) and thermal (Endo-Ice?) stimuli for 5 seconds. Dentin hypersensitivity was measured using a numerical scale. The aesthetic satisfaction and quality of life were evaluated by the patients and the OHIP-14 form, respectively. The gingival recession etiology was also investigated by anamnesis and clinical exam. All gingival defects were surgically treated by the coronally advanced flap technique combined with sub-epithelial connective tissue graft. All clinical parameters were evaluated at baseline and 3 months after treatment. The mean root coverage after 90 days was 67.90%. Complete root coverage was achieved in 44% of the treated teeth. ABSTRACT The aim of this study was to determine whether topical application of distilled spirits, cacha?a and whiskey, alter the epithelial thickness of tongue mucosa of hamsters. After sample size calculation, 720 microscopic fields were used to measure the total thickness of the tongue epithelium (epithelial layer over the stratum corneum). Forty hamsters were divided into two groups, depending on the application of the test substance in group 1 (cacha?a 48?gl) and group 2 (whiskey 40?gl), both evaluated in the experimental periods of 13 and 20 weeks. The contralateral side of the tongue, which has not received the test substance was used as control. After euthanasia, the tongues were dissected, fixed in 10% formalin solution, processed to obtain histological sections and were stained by hematoxylin and eosin (HE). For the morphometric analysis, three slides per specimen were evaluated. The thickness of the epithelium, both edges of the tongue, was measured with the aid of the program Motic Images Plus 2.0 (Motic ?). ABSTRACT There is still a lot of discussion about the pathophysiology of Peripheral Giant Cell Lesion (PGCL) and Central Giant Cell Lesion (CGCL). These lesions show distinct clinical features, although they have similar histological characteristics. Thus, immunohistochemical studies in PGCL and CGCL are being done to improve understanding these diseases. It has been reported that high level of FASN and angiogenesis are linked with tumors development. However, remains unknown whether these events are involved in the pathogenesis of LPCG and LCCG. The aim of this research was to study FASN expression and angiogenesis degree between PGCL and CGCL, in addition, verify the correlation between this variables. Thus, 13 cases of CGCL and 14 cases of PGCL were selected and examined by immunoexpression of FASN, CD34, CD105 and D2-40. The immunoexpression of FASN was assessed in components cells of lesions, followed by measurement of Microvassel Density (MVD) and Microvassel Area (MVA) for each selected sample. Data collected was submitted to descriptive analysis and followed by Mann Whitney test, ?t? test to independent samples and Person?s and Spearman?s correlation. The results of this study indicate that: (1) there are no differences in FASN immunoexpression between group lesions (MC ? 8% FASN+ / MGC ? 38% FASN+); (2) PGCL have greater MVD in CD34 than CGCL; there are no MDV differences in CD105 and D2-40 between lesions. ABSTRACT The aim of this study was to evaluate the influence of root canal treatment (RCT) quality, factors related to the coronal restoration (CR), carious lesions and periodontal clinical parameters in the periapical status of endodontically treated teeth. The patients were treated by undergraduate students at the Clinic of Endodontics, Federal University of Jequitinhonha and Mucuri Valleys (UFVJM) in the period between 2004-2009. The clinical and radiographic recall was performed between September 2010 and February 2011. Data were collected in records dentistry (age, gender, treated tooth, periapical status preoperative, type of treatment, instrumentation and obturation techniques, final radiography of the filling and the type of coronal restoration). Clinically, factors related to carious lesions, CR and periodontal parameters were considered. Radiographic parameters were evaluated the periapical status, carious lesions, CR and cast post. ABSTRACT The all-on-four concept presents high success rates of clinical follow-up studies. However,?different studies?select fully acrylic provisional bridges for?prosthetic initial?phase, allowing the incidence of mechanical complications from 11% to 30%. Studies reported the application of high technology to prepare the prosthetic framework such as CAD-CAM systems and intraoral welding, to minimize the failures. In an approach that uses known principles and has not added together in the same way, this study evaluated prosthesis with a metal framework without welding supported by prefabricated cylinders. Two acrylic prosthesis design, representative for the All-on-four, with a new configurations were selected for this study. Two control groups were produced G1 (negative control, n=10) and G4 (positive control, n=10). ABSTRACT Studies regarding the development of enamel defects and association with natal and postnatal conditions are scarce. Thus, the aim of this study was to evaluate the prevalence of enamel defects in primary teeth and determine whether prematurity, birth weight and sociodemographic variables may be associated with such defects. This was a cross-sectional study with 381 children aged between 3 and 5 years. Data were collected through clinical examination and interviews with parents during the National Immunization Day. The diagnosis of enamel defects was performed using the Development Defects of Enamel Index (DDE Index). During interviews, information were collected on sociodemographic aspects, pregnancy, birth weight, child prematurity and breastfeeding. Statistical analysis was performed using the SPSS software for Windows 17.0 and involved descriptive analysis, the Fisher?s test, qui-square test and Poisson regression. The prevalence of enamel defects was 29.9% and demarcated opacity was the most frequent type of defect. Children with low birth weight presented greater prevalence of enamel defects [PR (CI 95%) = 2,7 (1.66-4.61)]. In the first article, the authors reviewed the literature on cancer and propolis, addressing experimental carcinogenesis, chemically induced by 9,10-dimethyl-1,2-benzanthracene (DMBA), the chemical composition of propolis and its chemopreventive effects. The aim of the research paper was to evaluate the?tissue response?of tongue mucosa of hamsters, subjected to daily and alternating application of ethanolic extract of propolis (EEP) of a commercial presentation. ABSTRACT The present work describes the phytochemical and pharmacological investigation on extracts of the leaves and stems and flowers of the species Plectranthus neochilus Schltr. belonged to the family Lamiaceae. The plant is aromatic herb popularly known in Brazil as ?boldo? or ?boldo-gamb??, and as the species P. barbatus Andrews (?boldo-brasileiro") e P. ornatus Codd. ("boldinho"), according to ethnopharmacological survey, is often used in the treatment of hepatic insufficiency and dyspepsia, replacing the Peumus boldus Molina (?boldo-do-chile?). The phytochemical investigation of the hexanic extract of the P. neochilus leaves and stems yielded two triterpenes: ?-amyrin (PN1) e friedelin (PN2). The compound ?-amirin was isolated as a fatty acid ester in position C-3. In this extract was also obtained the mixture of the steroids sitosterol and stigmasterol (PN3). The phytochemical study conducted with the ethyl acetate extract of the leaves and stems gave the methoxy flavone at positions C-6 and C-7, the cirsimartin (PN4). All compounds obtained were first described in the species. Structural elucidation of isolated compounds was based on spectroscopic analysis of 1D NMR, HPLC, ESY and by comparison with data found in literature. ABSTRACT The present work presents simple, fast and low-cost methods for determination of sildenafil citrate (CS) and carbamazepine (CBZ) in pharmaceutical formulations using flow injection analysis (FIA) with multiple pulse amperometric (MPA) detection on the boron-doped diamond (DDB) working electrode. The method for the CS determination consists of the application of three sequential potential pulses as function of the time. SC is detected at 1.6 V and 1.9 V by two different irreversible oxidation processes. The third potential pulse (1.0 V) is applied for the regeneration (cleaning) of the electrode (boron-doped diamond) surface. The limit of detection (LD) in an order of 10 nmol L-1 was reached with a standard deviation of less than 0.2 % (n = 10) for 4.5 x 10-5 mol L-1 CS. The analytical frequency was calculated in 86 injections per hour. The addition-recovery studies in all the samples were around 100% and the results were validated by chromatographic methods. For the CBZ determination, two analytic conditions were proposed, one used for determination of a drug in pharmaceutical samples and other for detection the active ingredient in biological fluids. ABSTRACT Amylases are enzymes that act in the rupture of glycosidic bonds present in the chains of amylose and amylopectin. Amylases have applications in diverse areas such as pulp and paper industry, textiles, laundry detergents and cleaning products, chemical and pharmaceutical industry, production of vitamins and antibiotics. Although the amylases are found in plants, animals and micro-organisms, microbial enzymes generally meet satisfactorily the industrial demand. The microbial enzymes can be obtained by fermentative procedures, which may be obtained by cultivation both surface and submerged cultivation of micro-organisms. This study aimed to evaluate the production of amylolytic enzymes by submerged fermentation using Bacillus amyloliquefaciens ATCC 23350, and co-products from biodiesel production chain as substrates. The capacity of the production of amylolytic enzymes was tested using different substrates such as co-products, these being: macauba cake, palm kernel cake, peach palm flour, castor bean and lupine cake. The peach palm flour and macauba cake were the substrates which show greater production capacity of amylolytic enzymes, and co-products selected for the optimization step of enzyme production. ABSTRACT The search for alternative energies, sustainable and economically viable, highlighted the use of biofuels. Recent studies indicate the use of residual biomass as potential candidates for bioenergy production. In this context, the present dissertation aimed at evaluating the palm cake as a feedstock for the production of second generation ethanol through studies involving acid and alkaline pretreatment steps, saccharification and fermentation. Initially it was determined the chemical composition of palm cake, indicating the presence of 53.2% of total carbohydrates, of which 32% related to cellulose. To optimize the pretreatment was applied a Factorial Design by central composite rotational design. The program STATISTICA version 8.0 (Statsoft Inc., Tulsa,) was used for data analysis. Pretreatment with dilute sulfuric acid resulted in the removal of at least 82% of the hemicellulose present in palm cake. The subsequent alkaline pretreatment was able to remove at least 72% of the lignin from the biomass, an interfering of fermentation. ABSTRACT Electrodes comprised of ?-PbO2 were prepared by electrodeposition, by moans of the inclusion of Fe3+ and F- ions in the precursor mixture, using as substrates steel screen (ASTM 316) and carbon cloth in order to produce ozone in an filter-press electrochemical reactor conducted in the absence of liquid electrolyte. The analysis of SEM micrographs revealed that for both substrates at low dopants concentrations resulted in the formation of well-defined grains with a uniform surface area. X-rays analysis for both substrates showed the inhibition of formation of the ?-PbO2 phase for most of the dopant concentration. Cronopotenciometric studies revealed a good stability of the anodes when they were subjected to the intense gas evolution. For the electrode doped with 1 mM Fe3+, the current efficiency of EOP were 10% and 9.5% for an ozone generation rate of 1.35 g h-1 and 1.40 g h -1 using the steel screen and carbon cloth substrates, respectively. These results are promising, indicating a feasibility of using these electrodes for application in water purification, so the electrochemical ozone produced was applied to treat residuary water obtained from the biodiesel produced in the pilot plant of UFVJM. ABSTRACT The wine is one of the oldest beverage produced and consumed by humanity. In the last years, the research in food quality are looking for a new analytical techniques more precise, faster and accurate. In this way, the wineries demand for new analytical methods that can provide simultaneous data for a large number of analyses. The near infrared spectroscopy (NIR) is a very important analytical tool in wine industry, allowing the establishment of numerous analytical methodologies. In that way, the work developed for this dissertation aimed use near infrared spectroscopy and multivariate data analysis tools for the establishment of chemometric analysis of physicochemical quality of red wines. The wine samples was analyzed for 20 parameters using enological laboratorial methods and by using near infrared spectroscopy. The chemical an spectroscopic data was analyzed using different types of regression (multiple linear regression, principal components regression and least partial squares) and preprocessing tools to evaluate the correlation between the data. ABSTRACT In the first part of the proposed pattern recognition Jabuticaba (Myrciaria jabuticaba (vell) berg (jabuticaba sabar?)) for the evaluation of the elements boron, nitrogen, magnesium, phosphorus, potassium, calcium, manganese, iron, copper and zinc on the shell employing the techniques of titration, atomic absorption spectroscopy and molecular. The data obtained by analysis of the elements were arranged in a matrix 10 x 7, seven samples analyzed referring to the seven cities (S?o Jo?o da Chapada, Gouveia, Guinda, Couto de Magalhaes, Wells Caldas, Belo Horizonte and Serro) and ten variables corresponding concentrations of certain elements in MGK-1. Pattern recognition was done using the Analysis of Principal Components Analysis for Hierarchical Groups. This made it possible to identify the Principal Component Analysis groups formed by similarity, based on the weights corresponding to the influences of variables. ABSTRACT Pseudobrickellia brasiliensis (Spreng.) R.M. King & H. Rob (Asteraceae) is a species of Asteraceae popularly known as arnica-do-mato, arnica-do-campo or arnica simply. It is used in folk medicine against wounds and body aches, but there lack of scientific literature and found only one article on phytochemical study. Aiming to contribute to the study of P. brasiliensis, this work reports the phytochemical and biological tests that investigated the anti-inflammatory and antioxidant activity of extracts of the leaves. The leaves were collected in the city of Diamantina, in the Campus JK of UFVJM. Part of the leaves still fresh, were used for extraction of essential oil, and another part of the plant material was dried and subjected to extraction with solvents of different polarities - hexane, ethyl acetate, ethanol and water - resulting in four extracts. The constituents of the essential oil were identified by GC-MS and was found 25 terpenes, of which the major was the monoterpenes ?-thujene (17.21%) and ?-pinene (32.61%). ABSTRACT Coffee is among the most important agricultural products in international trade and its quality as a product is an important factor in price formation, besides being one of the most widely consumed beverages worldwide and therefore arouses interest for studies of the physiological effects of its constituents. ABSTRACT In this work, the development of an immunosensor for detection of anti-Leishmania infantum antibodies was performed. The principle of transduction used was the Surface Plasmon Resonance (SPR) and the sensor was constructed by covalent immobilization of soluble antigens of Leishmania infantum on self-assembled monolayers (SAMs) of 11-mercaptoundecanoic acid (11-MUA) which was activated by the mixture 100.0 mmol.L-1 1-ethyl-3 (3-dimethylaminopropyl) carbodiimide (EDC) and 150 mmol.L-1 N-hidroxisuccinimide (NHS). Cyclic Voltammetry and Dessortive Reduction by Differential Pulse Voltammetry techniques were employed in monitoring of SAM formation. Under optimized conditions, 1.0 mmol.L-1 of the 11-MUA in ethanolic solution and 24 hours were sufficient for the formation of a stable SAM on the gold surface. To obtain further information about the SAM, the Surface Coverage, Dielectric Constant and Thickness of the thiol film adsorbed on gold substrate were performed. ABSTRACT Myrciaria cauliflora (jabuticabeira) is a fruit specie, native in Brazil and created mainly in domestic gardens in Minas Gerais. However, little is known about its chemical composition and biological effects in the fruit residue. In front of this, the present work had as a goal to evaluate the chemical composition, antioxidant potential (in vitro) and hypolipidemic (in vivo) of Myrciaria cauliflora fruit peel flour (FCJ). For that, mature fruits (jabuticaba) were pulped manually and its peel was dried and crushed to obtain a homogeneous flour (FCJ). There had been evaluated the proximate composition, mineral content as well as total phenolic, flavonoids and anthocyanins as their antioxidant activity in vitro through different methods (DPPH, ABTS?+, FRAP and ?-caroten/acid linoleic). The FCJ was also subjected to extraction with water-alcohol extracts fractionated in column chromatography (CC). The phytochemicals isolated were characterized IR and NMR. ABSTRACT Nowadays, one cannot ignore the need for developing technologies that support the demands for renewable energy. One of the pathways for production of renewable energy is the transformation of vegetal biomass to liquid fuels. The agro-industrial residues as raw materials are potential candidates for this new energy industry. Thus the aim of the present dissertation was to evaluate the potential of cottonseed cake and crushed malt with peach palm flour as raw materials for the production of second generation ethanol and optimize step of acid pretreatment of lignocellulosic material. Initially was determined the chemical composition of cotton seed cake and crushed malt in relation to the percentages of moisture, ash, ether extract, crude protein, total soluble sugars, starch, hemicellulose, cellulose, lignin, neutral detergent fiber and acid detergent fiber. With the results of chemical characterization it was possible to estimate a potential yield of more than 300L of ethanol / ton of each residue. For the optimization of acid pretreatment for hemicelluloses removal was applied a Central Composite Design. The software STATISTICA Version 8.0 (Statsoft Inc., Tulsa,) was used for data analysis. The goodness of fit of the model equation was expressed by the coefficient of determination (R?) and its statistical significance conditioned by F-test. The insoluble material recovered after the optimized pretreatment had starch and hemicellulose fractions completely removed. The hydrolyzate obtained under optimized conditions of pretreatment showed 15.7% of reducing sugars to the crushed malt and 5.9% of reducing sugars for cottonseed cake. The treatment of the hydrolysates with activated charcoal removed no less than 58% of the phenolic compounds present there. Fermentability tests with Saccharomyces cerevisiae and Pichia stipitis have been promising. Fermentation with S. cerevisiae showed a YP/S of 0.30 for cottonseed cake and 0.37 for the crushed malt. Fermentative yields with P. stipitis ranged from 9% to 25% for agro-industrial residues.
