We developed cationic liposomes containing DNA through a conventional process involving steps of (i) preformation of liposomes, (ii) extrusion, (iii) drying and rehydration and (iv) DNA complexation. Owing to its high prophylactic potentiality against tuberculosis, which had already been demonstrated in preclinical assays, we introduced modifications into the conventional process towards getting a simpler and more economical process for further scale-up. Elimination of the extrusion step, increasing the lipid concentration (from 16 to 64 mM) of the preformed liposomes and using good manufacturing practice bulk lipids (96-98% purity) instead of analytical grade purity lipids (99.9-100%) were the modifications studied. The differences in the physico-chemical properties, such as average diameter, zeta potential, melting point and morphology of the liposomes prepared through the modified process, were not as significant for the biological properties, such as DNA loading on the cationic liposomes, and effective immune response in mice after immunisation as the control liposomes prepared through the conventional process. In this paper, Co/CeO2 catalysts, with different cobalt contents were prepared by the polymeric precursor method and were evaluated for the steam reforming of ethanol. The catalysts were characterized by N-2 physisorption (BET method), X-ray diffraction (XRD), UV-visible diffuse reflectance, temperature programmed reduction analysis (TPR) and field emission scanning electron microscopy (FEG-SEM). It was observed that the catalytic behavior could be influenced by the experimental conditions and the nature of the catalyst employed. Physical-chemical characterizations revealed that the cobalt content of the catalyst influences the metal-support interaction which results in distinct catalyst performances. The catalyst with the highest cobalt content showed the best performance among the catalysts tested, exhibiting complete ethanol conversion, hydrogen selectivity close to 66% and good stability at a reaction temperature of 600 degrees C. This paper proposes three new hybrid mechanisms for the scheduling of grid tasks, which integrate reactive and proactive approaches. They differ by the scheduler used to define the initial schedule of an application and by the scheduler used to reschedule the application. The mechanisms are compared to reactive and proactive mechanisms. Results show that hybrid approach produces performance close to that of the reactive mechanisms, but demanding less migrations.

Persistent organic pollutants (POPS) present in the living environment are thought to have detrimental health effects on the population, with pregnant women and the developing foetus being at highest risk. We report on the levels of selected POPs in maternal blood of 155 delivering women residing in seven regions within the Sao Paulo State, Brazil. The following selected POPs were measured in the maternal whole blood: 12 polychlorinated biphenyls (PCBs) congeners (IUPAC Nos. 99, 101, 118, 138, 153, 156, 163, 170, 180, 183, 187, 194); dichlordiphenyltrichloroethane p,p'-DDT, diphenyldichloroethylene p,p'-DDE and other pesticides such as hexachlorocyclohexanes (alpha-HCH, beta-HCH, gamma-HCH), hexachlorobenzene (HCB), chlordane derivatives cis-chlordane, trans-chlordane. oxy-chlordane, cis-nonachlor and trans-nonachlor. Statistical comparisons between regions were performed only on compounds having concentrations above LOD in 70% of the samples. PCB118 congener was found to be highest in the industrial site (mean 4.97 ng/g lipids); PCB138 congener concentration was highest in the Urban 3 site (mean 4.27 ng/g lipids) and congener PCB153 was highest in the industrial and Urban 3 sites with mean concentration of 7.2 ng/g lipids and 5.89 ng/g lipids respectively. In this paper we report original measurements of total cross sections (TCSs) for positron scattering from the cyclic ethers oxirane (C2H4O), 1,4-dioxane (C4H8O2), and tetrahydropyran (C5H10O). The present experiments focus on the low energy range from similar to 0.2 to 50 eV, with an energy resolution smaller than 300 meV. This study concludes our systematic investigation into TCSs for a class of organic compounds that can be thought of as sub-units or moieties to the nucleotides in living matter, and which as a consequence have become topical for scientists seeking to simulate particle tracks in matter. Note that as TCSs specify the mean free path between collisions in such simulations, they have enjoyed something of a recent renaissance in interest because of that application. For oxirane, we also report original Schwinger multichannel elastic integral cross section (ICS) calculations at the static and static plus polarisation levels, and with and without Born-closure that attempts to account for the permanent dipole moment of C2H4O. Those elastic ICSs are computed for the energy range 0.5-10 eV. To the best of our knowledge, there are no other experimental results or theoretical calculations against which we can compare the present positron TCSs. However...

Aim. To evaluate the psychometric Instrument to Measure the Impact of Coronary Disease on Patients Everyday Life (IDCV) when applied on patients with coronary disease being followed at an outpatient clinic. Methods. One hundred and fifty-three patients with coronary disease were registered. Patients were analysed in terms of acceptability, ceiling and floor effect, reliability (using Cronbach's alpha coefficient) and convergent construct validity (by means of Spearman correlation) between the domains of IDCV and of ` Medical Outcomes Trust Short-form Health Survey (SF-36)' and ` MacNew Heart Disease Health-related Quality of Life Questionnaire'. Results. Evidence was found for high acceptability of IDCV. No evidence was found for ceiling and floor effects regarding the total score of IDCV; however, ceiling and floor effects were found from the adjustment to the disease domain. Evidence was found for reliability of the instrument as a whole and for its domains (Cronbach's a ranged between 0 70-0 85). Objective: The aim of this study was to analyse the medical indication and the use of elastic compression stockings, and to assess patient adherence to treatment in different regions of Brazil. Method: The prescription and clinical indication of elastic stockings were evaluated in a prospective, descriptive, cross-sectional, multicentre study for a population of private patients. In 2009, 3414 patients from 123 treatment centres in southern, south-eastern and north-eastern Brazil were evaluated using a questionnaire. The following variables were analysed: sociodemographic (gender, age, occupation and education), lifestyle (physical activity and time spent standing); classification of venous disease (CEAP [clinical, aetiological, anatomical and pathophysiological] classification - clinical criteria), indications for prescription, consumer behaviour (strength, acquisition and use of stockings) and criteria of satisfaction (improvement, duration of use, adherence). The effects of compression therapy were assessed at a follow-up visit approximately 30 days after starting treatment with the following items being assessed: complaints about pain, discomfort, burning sensation and oedema of the leg while using elastic stockings. Multivariate analysis was used to compare data with an alpha error of 5% (P value < 0.05) being considered acceptable. Results: The average age increased with the severity of chronic venous insufficiency; the main indications used by physicians were leg pain and discomfort; 89.3% of patients bought stockings and thus started treatment with more than 90% of these reporting improvements in symptoms. A proposta deste estudo é desvelar o sentido de Ser-criança com câncer em tratamento ambulatorial, utilizando a brinquedo-teca como possibilidade de favorecer a expressão, pela criança, de seu mundo cotidiano. Participaram sete crianças entre três e nove anos, com diagnóstico de algum tipo de câncer infantil. A fim de desvelar o sentido das vivências das crianças com câncer, foi realizada uma análise à luz da fenomenologia existencial de Martin Heidegger. A criança-com-câncer configurou-se como um ir e vir permeado ora pela autenticidade, quando a criança assumia sua doença e seu ser-para-a-morte, ora pela inautenticidade, quando se deixava levar pelo modo de ser da decadência dos familiares e da equipe de saúde. O brincar pôde favorecer um rico acesso às vivências da criança gravemente doente.; The purpose of this study is to learn the meaning of being-a-child with cancer submitted to ambulatory treatment and using the toy library as a possibility to enable their expression about their world. The water activity of aqueous solutions of EO-PO block copolymers of six different molar masses and EO/PO ratios and of maltodextrins of three different molar masses was determined at 298.15 K. The results showed that these aqueous solutions present a negative deviation from Raoult's law. The Flory-Huggins and UNIFAC excess Gibbs energy models were employed to model the experimental data. While a good agreement was obtained with the Flory-Huggins equation, discrepancies were observed when predicting the experimental behavior with the UNIFAC model. Hábitos de vida saudáveis e uma dieta balanceada aliados ao alto consumo de frutas e vegetais estão associados à redução do risco de doenças e à manutenção da saúde. O óleo de abacate possui em sua composição substâncias bioativas capazes de prevenir e controlar as dislipidemias. Como existem poucas pesquisas científicas avaliando o potencial deste óleo para o consumo humano, o presente trabalho estudou os processos de extração e refino do óleo de abacate. Para tanto, procedeu-se à separação da polpa das outras partes da fruta. A polpa fresca foi seca em estufa, e posteriormente moída para a obtenção de um farelo. O óleo obtido do farelo foi extraído e caracterizado. Os resultados mostraram que os processos de extração e refino do óleo a partir da variedade Margarida são tecnicamente viáveis, o que o torna excelente matéria-prima para a indústria alimentícia. Além disso, possui um perfil de ácidos graxos e esteróis muito semelhante ao perfil do azeite de oliva, podendo desta forma, substituir o óleo de soja e ser utilizado juntamente com o azeite de oliva nos óleos mistos, oferecendo ao consumidor brasileiro um produto de qualidade superior e com menor custo.; Healthy life habits and an adequate diet...

The enzyme UDP-N-acetylglucosamine pyrophosphorylase (PyroMp) from Moniliophthora perniciosa (CCMB 0257), a pathogenic fungal strain and the causative agent of the witches' broom disease in Theobroma cacao, was partially purified by precipitation with ammonium sulfate and gel filtration on Sephacryl S-200. The buffer for enzyme extraction was sodium phosphate, 0.050 mol L-1, pH 7.0, containing 1.0 mol L-1 NaCl. Response surface methodology (RSM) was used to determine the optimum pH and temperature conditions. Four different isoenzymes (PyroMp I, PyroMp II, PyroMp III and PyroMp IV) were obtained with optimal pH ranging from 6.9-8.4 and optimum temperature ranging from 28 to 68 °C. The 3D structure of pyrophosphorylase of M. perniciosa was determined by comparative modeling. The model obtained showed a good quality, possessing 78.6% of amino acids in energetically allowed regions. The model was then submitted for DM simulation and showed a good geometric quality (91.1% Ramachandran plot). The active site of the enzyme was found to be extremely well conserved. O inventário florístico de um fragmento de floresta semidecídua foi conduzido durante o período de maio de 1996 a abril de 1998 em um hectare de floresta localizado na região central da Reserva de Santa Genebra (22º49'45" S e 47º06'33" W) a 580-610 m de altitude. Foram identificadas 201 espécies, distribuídas em 57 famílias e 147 gêneros. Fabaceae e Rubiaceae (18 espécies), Myrtaceae (14 spp.), Rutaceae (11 spp.), Solanaceae e Sapindaceae (nove spp.), Bignoniaceae, Meliaceae e Euphorbiaceae (oito spp.), Malvaceae (sete spp.) foram as famílias mais ricas. Cerca de 70% das espécies foram classificadas como secundárias tardias e secundárias tardias de sub-bosque. A comparação de espécies arbóreas com 25 fragmentos do Estado de São Paulo e Norte do Paraná mostraram que a flora da reserva é mais similar às florestas da região de Campinas. Objetivou-se, neste trabalho, avaliar a produção e qualidade de botões de rosa cultivar Vega ( Rosa sp. ), cultivada sob cobertura plástica com diferentes anos de uso. Para tanto foram selecionados 15 vãos de estufas com plásticos colocados em 1998, 1999, 2002, 2003, e testemunha (sem uso anterior), com três repetições, avaliados por duas maneiras: na primeira os frutos foram colhidos através de uma poda (como procedimento do cultivo comercial); na segunda foi realizada uma poda rasa em uma área de 1 m² para poder acompanhar o crescimento da haste floral desde o início da formação até a fase de botão floral. No primeiro caso foram realizadas 4 coletas ao longo do período do experimento, nas quais foram coletadas 4 hastes florais por tratamento, totalizando 60 flores por data de coleta. No segundo caso foi avaliado o aparecimento dos botões florais dos novos brotos em quatro datas até a formação das hastes florais comerciais, e então coletadas 13 hastes florais por tratamento. Não houve diferença estatística entre os anos de uso dos plásticos de 1998, 1999, 2002 e 2003 nas variáveis: comprimento, diâmetro e comprimento dos botões florais, massa fresca e seca das pétalas, para as condições e cultivar analisados. Brazilian exports of fresh citrus represent less than 1% of the overall Brazilian production. Phytosanitary barriers and poor appearance stand out among the several reasons contributing to such low exporting/production ratio. The purpose of this work was to characterize postharvest injuries in 'Valencia' sweet oranges and 'Murcott' tangors produced for foreign markets after different processing stages in a packinghouse, as well as to identify critical points and impact extent on packing lines. Sampling was performed both after pre-washing and degreening the fruits, and also at the arrival on the packing table and in the pallet. They were stored for 21 days at 25ºC and 85% RH. The incidence of injuries was visually assessed every three days. An instrumented sphere with acceleration register was used to evaluate the extent of impacts (G, m/s²) at the 19 transference points of the citrus processing line. There was low rot incidence (under 3.5%) at the different stages of fruit processing, with slight increase after degreening in 'Valencia' orange and a decrease after fungicides treatment in 'Murcott' tangor. The main pathogens found in the oranges were Lasiodiplodia theobromae and Penicillium digitatum, which were surpassed by Colletotrichum gloeosporioides in the tangors. A wide array of molecular markers has been used to investigate the genetic diversity among common bean species. However, the best combination of markers for studying such diversity among common bean cultivars has yet to be determined. Few reports have examined the genetic diversity of the carioca bean, commercially one of the most important common beans in Brazil. In this study, we examined the usefulness of two molecular marker systems (simple sequence repeats - SSRs and amplified fragment length polymorphisms - AFLPs) for assessing the genetic diversity of carioca beans. The amount of information provided by Roger's modified genetic distance was used to analyze SSR data and Jaccards similarity coefficient was used for AFLP data. Seventy SSRs were polymorphic and 20 AFLP primer combinations produced 635 polymorphic bands. Molecular analysis showed that carioca genotypes were quite diverse. AFLPs revealed greater genetic differentiation and variation within the carioca genotypes (Gst = 98% and Fst = 0.83, respectively) than SSRs and provided better resolution for clustering the carioca genotypes. SSRs and AFLPs were both suitable for assessing the genetic diversity of Brazilian carioca genotypes since the number of markers used in each system provided a low coefficient of variation. ‣ Genetic diversity in Brazilian sweet potato (Ipomoea batatas (L.) Lam., Solanales, Convolvulaceae) landraces assessed with microsatellite markers

We used simple sequence repeat (SSR) markers to investigate the genetic diversity of 78 sweet potato (Ipomoea batatas) accessions (58 landraces and 20 putative clones) from traditional agricultural households from 19 local communities in the Vale do Ribeira, São Paulo, Brazil. Eight SSR loci were assessed using 6% (w/v) polyacrylamide gels stained with silver nitrate and the accessions genotyped considering the presence or absence of bands. The results were subjected to analysis of molecular variance (AMOVA), and cluster and principal coordinate analyses. Spatial structure was assessed using Mantel's test to compare genetic and geographic distances. Each primer pair generated between three and ten clearly scorable polymorphic fragments. Cluster analyses showed a Jaccard's index from 0.3 to 1.0, indicating high genetic and intravarietal diversity. Accessions from all 19 communities were not spatially structured (r = 0.15, p < 0.054), with AMOVA indicating that most of the variability (58.2%) was distributed within households and only 18.1% of the variability was distributed between households within communities. ‣ Phosphorus availability of rock phosphates as compared with feed-grade phosphates for swine

One hundred ninety two swine were used in a trial to assess the relative bioavailability of phosphorus (RBP) in six phosphate sources. Phosphates were three feed grade phosphates (FP), two made in Brasil, and one USA made, and three rock phosphate samples (RP) originated from two mines sites in Brasil, and one mine site in Israel. Levels of calcium, phosphorus and fluorine in RP were 29, 12 and 1.7% (RP source 1), 33, 14 and 1.4% (RP source 2), and 30, 14 and 3.6% (RP source 3), respectively. Pigs were fed a corn-soybean meal basal diet (18% CP, 0.95% Lys, 0.75% Ca, 0.37% P) or the basal diet with 0.15% P from a standard purified grade calcium phosphate (SP), or with 0.15% P from experimental FP or RP. Each diet was fed to six pen replicates of four pigs per pen for 35 days (14.4 to 39.9 kg). Weight gain (WG), feed/gain (FG), plasma P (PP), bone ash (BA), and breaking strength of metacarpals and metatarsals (BS-MM) and femurs (BS-F) were improved by phosphorus addition. However, performance and bone parameters were depressed by RP, as compared to FP dietary supplementation. O sudoeste paranaense localiza-se na transição entre a Floresta Estacional Semidecidual (FES) e a Floresta Ombrófila Mista (FOM), numa região de grande relevância ecológica, por conter duas importantes formações florestais do sul-sudeste brasileiro. Entretanto, essa mesma região é marcada pela escassez de levantamentos florísticos e de caracterização da vegetação. Nesse trabalho é feita a caracterização florística e estrutural de três áreas de vegetação nativa, localizadas em Quedas do Iguaçu, sudoeste do Paraná, na bacia do rio Iguaçu. Além disso, foi feita a caracterização fitogeográfica das florestas estudadas, com base nos dados coletados e na listagem de espécies amostradas em outras 52 áreas de FES ou FOM do Brasil. Em cada uma das três áreas estudadas, a vegetação arbustivo-arbórea (diâmetro à altura do peito > 5 cm) foi caracterizada por meio da alocação de pontos quadrantes. No total foram encontradas 128 espécies, com variação entre 63 a 78 para cada área. Cada área apresentou distinção quanto à estrutura e às espécies mais abundantes.
