ABE, V.C.

The roles that nitric oxide (NO) plays in the cardiovascular system of reptiles are reviewed, with particular emphasis on its effects on central vascular blood flows in the systemic and pulmonary circulations. New data is presented that describes the effects on hemodynamic variables in varanid lizards of exogenously administered NO via the nitric oxide donor sodium nitroprusside (SNP) and, preliminary data on the effects of SNP inhibition of nitric oxide synthase (NOS) by L-nitroarginine methyl ester (L-NAME). Furthermore. on hemodynamic variables in the tegu lizard are presented. The findings are compared with previously published data from Our laboratory on three other species of reptiles: pythons (Skovgaard, N., Galli, G., Taylor, E.W., Conlon, J.M., Wang.. T., 2005. Hemodynamic effects of python neuropeptide gamma in the anesthetized python, Python regius. Regul. Pept. 18, 15-26), rattlesnakes (Galli, G., Skovgaard, N., Abe, A.S., Taylor, E.W., Wang, T., 2005. Transcriptional control of the low-temperature-inducible icdII gene, encoding the thermolabile isocitrate dehydrogenase of a psychrophilic bacterium, Vibrio sp. strain ABE-1, was found to be mediated in part by a transcriptional silencer locating at nucleotide positions −560 to −526 upstream from the transcription start site of icdII. Deletion of the silencer resulted in a 20-fold-increased level of expression of the gene at low temperature (15°C) but not at high temperature (37°C). In addition, a CCAAT sequence located 2 bases upstream of the −35 region was found to be essential for the low-temperature-inducible expression of the gene. By deletion of this sequence, low-temperature-dependent expression of the gene was completely abolished. The ability of the icdII promoter to control the expression of other genes was confirmed by using a fusion gene containing the icdII promoter region and the promoterless icdI open reading frame, which encodes the non-cold-inducible isocitrate dehydrogenase isozyme of Vibrio sp. strain ABE-1. Escherichia coli transformants harboring icdII acquired an ability to grow rapidly at low temperature.

Four mutants of the virulent Mahoney strain of poliovirus were generated by introducing mutations in nucleotides (nt) 128 to 134 of the genome, a region that contains a part of the stem-loop II (SLII) structure located within the internal ribosomal entry site (IRES; nt 120 to 590) (K. Shiroki, T. Ishii, T. Aoki, Y. Ota, W.-X. Yang, T. Komatsu, Y. Ami, M. Arita, S. Abe, S. Hashizume, and A. Nomoto, J. Virol. 71:1–8, 1997). These mutants (SLII mutants) replicated well in human HeLa cells but not in mouse TgSVA cells that had been established from the kidney of a poliovirus-sensitive transgenic mouse. Their neurovirulence in mice was also greatly attenuated compared to that of the parental virus. The poor replication activity of the SLII mutants in TgSVA cells appeared to be attributable to reduced activity of the IRES. Two and three naturally occurring revertants that replicated well in TgSVA cells were isolated from mutants SLII-1 and SLII-5, respectively. The revertants recovered IRES activity in a cell-free translation system from TgSVA cells and returned to a neurovirulent phenotype like that of the Mahoney strain in mice. Two of the revertant sites that affected the phenotype were identified as being at nt 107 and within a region from nt 120 to 161. A mutation at nt 107...

The expression of two structurally different isocitrate dehydrogenase isozymes of Vibrio sp. strain ABE-1 in Escherichia coli was examined. At a low temperature (15 degrees C), a thermolabile and monomeric type isozyme (IDH-II), which is quite different in amino acid sequence from the E. coli isocitrate dehydrogenase, was expressed and conferred glutamate prototrophic ability on an E. coli mutant defective in isocitrate dehydrogenase. The ability of IDH-II to confer restoration of the E. coli mutant to glutamate prototrophy was similar to that of IDH-I, which is a dimeric enzyme homologous to the E. coli isocitrate dehydrogenase. At a high temperature (37 degrees C), no functional IDH-II was expressed. Transcription of icdI and icdII genes, which encode IDH-I and IDH-II, respectively, was regulated differently by different environmental conditions. The level of icdII mRNA was increased by lowering the growth temperature for E. coli transformants, while the level of icdI mRNA was increased when E. coli transformants were cultured in acetate minimal medium. Similar patterns of transcriptional regulation of the two icd gene were observed also in Vibrio sp. strain ABE-1. However, activity of isocitrate dehydrogenase kinase, which can phosphorylate IDH-I and consequently inactivate the enzymatic activity...

Vibrio sp. strain ABE-1 was grown in a medium that contained as its stable isotope tracer either [2,2-2H2]cis-9-hexadecenoic or [2,2-2H2]trans-9-hexadecenoic acid. Gas chromatographic-mass spectrometric analysis of the cis-9-hexadecenoic and trans-9-hexadecenoic acid fractions from the cells revealed the formation of an intracellularly isomerized 2,2-2H2-fatty acid which differed from the tracer only in the geometrical configuration of the double bond. This observation shows that cis-trans isomerization without a shift in double-bond position between these two geometric hexadecenoic acid isomers can occur in the cells.

The genes coding for two structurally different isocitrate dehydrogenase isozymes (IDH-I and IDH-II) of a psychrophilic bacterium, Vibrio sp. strain ABE-1, were cloned and sequenced. Open reading frames of the genes (icdI and icdII) are 1,248 and 2,229 bp in length, respectively. The amino acid sequences predicted from the open reading frames of icdI and icdII corresponded to the N-terminal amino acid sequences of the purified IDH-I and IDH-II, respectively. No homology was found between the deduced amino acid sequences of the isozymes; however, the IDH-I, a dimeric enzyme, had a high amino acid sequence identity (74.3%) to the Escherichia coli IDH. The deduced amino acid sequence of the IDH-II, a monomeric enzyme, was not related to any known sequence. However, the IDH-II had an amino acid sequence homologous to that of a cyanogen bromide-cleaved peptide containing a putative active-site methionyl residue of the monomeric IDH of Azotobacter vinelandii. The two genes (icdlI and icdII) were found to be tandemly located in the same orientation. Northern (RNA) blot analyses showed that the two genes are transcribed independently. Primer extension experiments located single transcriptional start sites 39 and 96 bp upstream of the start codons of icdI and icdII...

A high level of a trans-unsaturated fatty acid was found in the phospholipids of a psychrophilic bacterium, Vibrio sp. strain ABE-1. This fatty acid was identified as 9-trans-hexadecenoic acid (C16:19t) by gas-liquid chromatography and infrared absorption spectrometry. C16:1(9)t accounted for less than 1% of the total fatty acids in cells grown at 5 degrees C and reached 12% of the total at 20 degrees C. We suggest that the increase in the level of the trans-unsaturated fatty acid is related to the high growth rate of this bacterium at elevated temperatures. Possible biological roles of the trans-unsaturated fatty acid in the adaptation of the microorganism to the ambient temperature are discussed.

Sporulation of Clostridium botulinum types A, B, and E, Clostridium perfringens, and Putrefactive Anaerobe 3679 in dialysis sacs. J. Bacteriol. 85:126–133. 1963.—Concentrated cultures of spores of Clostridium botulinum type A (33A, 37A), B (41B, 51B), and E (strain VH), C. perfringens (strain E), and Putrefactive Anaerobe 3679 were prepared in intussuscepted cellulose dialysis tubing. The apparatus consisted of a telescoped cellulose bag immersed into a suitable sporulation medium in a large Pyrex tube. The initial inoculum was a heavy suspension in physiological saline solution of either vegetative cells or heat-shocked spores. The seed material was introduced into the interior of the dialysis bag. Maximal spore populations were obtained within 10 to 12 days. Strains of C. botulinum type E and C. perfringens, known for their poor sporulation in conventional cultures, gave good spore crops in the dialysis bag. Some crops were of the order of 1010 and 1011 viable spores per liter of medium. The spores produced in the dialysis bag were conspicuously large, particularly after incubation for 20 to 30 days. Observations of the characteristics of spores formed in telescoped bags indicate that two highly resistant strains of C. botulinum...

Syngeneic SL mice inoculated with murine myeloid leukemia cells (M1) all died of leukemia within 30 days. Treatment three times a week with 12.5-50 pmol per mouse of either 1 alpha,25-dihydroxyvitamin D3 [1 alpha,25(OH)2D3], the active form of vitamin D3, or its synthetic analog, 1 alpha-hydroxyvitamin D3 [1 alpha(OH)D3], considerably prolonged the survival time of mice inoculated with M1 cells. 1 alpha(OH)D3 was more effective than 1 alpha,25(OH)2D3 in increasing the survival time of the mice. 1 alpha(OH)D3 also increased the survival time of nude mice inoculated with M1 cells. The 1 alpha(OH)[3H]D3 administered intraperitoneally to tumor-bearing mice was converted very rapidly to 1 alpha,25(OH)2-[3H]D3. The chronic administration of 25 pmol of 1 alpha(OH)D3 to tumor-bearing mice for 30 days caused no appreciable hypercalcemia. These results indicate clearly that 1 alpha,25(OH)2D3 is effective not only in inducing differentiation of M1 cells in vitro, as previously reported [Abe, E., Miyaura, C., Sakagami, H., Takeda, M., Konno, K., Yamazaki, T., Yoshiki, S. & Suda, T. (1981) Proc. Natl. Acad. Sci. USA 78, 4990-4994], but also in prolonging the survival time of mice inoculated with M1 cells.

BALB/c mice bearing the transplanted lymphoid tumour ABE-8 developed increased levels of neutrophil, macrophage and eosinophil progenitor cells in the marrow and spleen. Neutrophil-macrophage progenitors, assayed as granulocyte-macrophage colony-forming cells in agar, increased in number only if invasion of the marrow or spleen by ABE-8 cells was demonstrable. Rises in B-lymphocyte colony-forming cells occurred whether or not there was invasion of the host spleen or marrow. No increase in progenitor cells was found in mice bearing diffusion chambers containing ABE-8 cells. The magnitude of leukaemic infiltration was determined by assaying numbers of leukaemic stem cells in the spleen and marrow using a selective cloning system. This transplanted lymphoid leukaemia appears to be a useful model for analysing the effects of haemopoietic tumours on host haemopoietic tissues.

The note describes the major findings of the study "Engaging with adults", which reviews the international experience with adult basic education (ABE), with a focus on Africa. As a foundation for improved conditions of life, basic education serves not only the young, but adolescents and adults who may have been missed by the educational system. Illiteracy is a major barrier to poverty alleviation, and needs to be addressed, through quality education, and effective ABE programs. The note outlines a range of policy issues for governments to strengthen its support of ABE, from targeting vulnerable groups, through the roles governments and nongovernmental organizations can play - including those of businesses and industry - to institutionalizing ABE, thus enhancing literacy and numeracy, as well as information technology. Recommendations suggest strong political leadership, and considerable investment in institutional development to diversify programs so as to be responsive to local demand.

Recently Abe (arXiv:cond-mat/1005.5110v1) claimed that the q-entropy of nonextensive statistical mechanics cannot be generalized for the continuous variables and therefore can be used only in the discrete case. In this letter, we show that the discrete q-entropy can be generalized to continuous variables exactly in the same manner as Boltzmann-Gibbs entropy, contrary to the claim by Abe, so that q-entropy can be used with discrete as well as continuous variables.; Comment: 6 pages, no figures

Topological excitations are usually classified by the $n$th homotopy group $\pi_n$. However, for topological excitations that coexist with vortices, there are case in which an element of $\pi_n$ cannot properly describe the charge of a topological excitation due to the influence of the vortices. This is because an element of $\pi_n$ corresponding to the charge of a topological excitation may change when the topological excitation circumnavigates a vortex. This phenomenon is referred to as the action of $\pi_1$ on $\pi_n$. In this paper, we show that topological excitations coexisting with vortices are classified by the Abe homotopy group $\kappa_n$. The $n$th Abe homotopy group $\kappa_n$ is defined as a semi-direct product of $\pi_1$ and $\pi_n$. In this framework, the action of $\pi_1$ on $\pi_n$ is understood as originating from noncommutativity between $\pi_1$ and $\pi_n$. We show that a physical charge of a topological excitation can be described in terms of the conjugacy class of the Abe homotopy group. Moreover, the Abe homotopy group naturally describes vortex-pair creation and annihilation processes, which also influence topological excitations. We calculate the influence of vortices on topological excitations for the case in which the order parameter manifold is $S^n/K$...

We address the problem of clustering words (or constructing a thesaurus) based on co-occurrence data, and using the acquired word classes to improve the accuracy of syntactic disambiguation. We view this problem as that of estimating a joint probability distribution specifying the joint probabilities of word pairs, such as noun verb pairs. We propose an efficient algorithm based on the Minimum Description Length (MDL) principle for estimating such a probability distribution. Our method is a natural extension of those proposed in (Brown et al 92) and (Li & Abe 96), and overcomes their drawbacks while retaining their advantages. We then combined this clustering method with the disambiguation method of (Li & Abe 95) to derive a disambiguation method that makes use of both automatically constructed thesauruses and a hand-made thesaurus. The overall disambiguation accuracy achieved by our method is 85.2%, which compares favorably against the accuracy (82.4%) obtained by the state-of-the-art disambiguation method of (Brill & Resnik 94).; Comment: latex file, uses colacl.sty file and 4 eps files, to appear in Proc. of COLING-ACL'98, 8 pages

Bilinear groups are often used to create Attribute-Based Encryption (ABE) algorithms. In particular, they have been used to create an ABE system with multi authorities, but limited to the ciphertext-policy instance. Here, for the first time, we propose two multi-authority key-policy ABE systems. In our first proposal, the authorities may be set up in any moment and without any coordination. A party can simply act as an ABE authority by creating its own public parameters and issuing private keys to the users. A user can thus encrypt data choosing both a set of attributes and a set of trusted authorities, maintaining full control unless all his chosen authorities collude against him. In our second system, the authorities are allowed to collaborate to achieve shorter keys and parameters, enhancing the efficiency of encryption and decryption. We prove our systems secure under algebraic assumptions on the bilinear groups: the bilinear Diffie-Hellmann assumption and an original variation of the former.; Comment: 32 pages
