Amorbus obscuricornis (Westwood) and Gelonus tasmanicus (Le Guillou) (Heteroptera: Coreidae) are specific to Eucalyptus (Myrtaceae). A obscuricornis feeds almost exclusively upon apical shoots and causes a characteristic wilting and necrosis. By comparison, the feeding activities of G. tasmanicus result in no obvious phytotoxicosis. Salivary gland extracts from both species exhibited sucrase activity but no pectinmethylesterase (PME) activity. Saliva from A obscuricornis also exhibited considerable oxidase activity. Sucrase activity was significantly higher in extracts derived from G. tasmanicus than from A. obscuricornis, but this could not explain the observed differences in phytotoxic symptoms. It is suggested that differences in plant damage are attributable to the site of feeding activity (i.e. young versus mature tissue), which predetermines the reactivity of host tissues, and/or the quantity of salivary enzymes injected.

Amongst the many variables that can determine success of cloning, the source of nuclei, the procedure used for nuclear transfer, and the activation of the reconstructed embryo are very important aspects. In this study, we have compared the two most common procedures for transferring nuclei to enucleated oocytes - cell fusion (CF) and piezoelectric microinjection (PEM) using different somatic cells - and we have investigated the effect of different activation procedures. Granulosa cells and fibroblasts were grown to confluency or in low serum to induce a quiescent state, while lymphocytes were thawed immediately prior to use. Enucleated oocytes were reconstructed either with CF or PME by 21-23 h postmaturation. For cell fusion, one pulse of 1 kVolt/cm for 30 μsec was used; for PEM, the cell membrane was broken by repeated pipetting and transferred in a 12% PVP solution to facilitate injection. Manipulated oocytes were activated with ionomycin and cycloheximide (CHX) or 6-DMAP (DMAP) and cultured in microdrops of SOF-BSA-AA. On day 7 (day 0: nuclear transfer), embryo development was evaluated and embryos were either transferred fresh or were frozen. Solubilization, transport and re-precipitation of Au in the supergene environment can lead to the formation of secondary Au enrichment zones, revealed as geochemical anomalies and secondary gold grains. Cyanide-producing microorganisms can contribute to the solubilization of Au, and the cyanide produced may play an important role in the formation of ‘bacterioform’ structures observed on gold grains. To examine the effect of bacterial cyanide production on Au surfaces, ultra-flat Au foil was incubated in peptone meat extract (PME) medium in the presence of the cyanide-producing bacterium Chromobacterium violaceum for up to 56 days. Within seven days C. violaceum had formed biofilms on the Au surfaces, and cyanide produced by the bacteria had a measurable effect on the Au surface compared to abiotic controls. After removal of the biofilm by sonication in de-ionized water, the surfaces incubated with C. violaceum were significantly rougher compared to the negative controls. This was demonstrated by root mean square roughness (RMS) analyses and statistical t-testing following scanning tunneling microscopy (STM). Approved for public release; distribution in unlimited.; This thesis analyzes the perceptions of a non-random sample of 210 officers and enlisted Marines in two locations. A researcher-developed survey and semi-structured interviews were administered to ascertain opinions of Marines concerning leadership development. An analysis of the content of leadership training and education courses was also conducted. This information was compared to contemporary leadership theory and relevant models of leadership. In general, leadership development provided is adequate, but is lacking in some areas of skill development, application of skills and values, and relevancy to contemporary leadership issues. Professional Military Education (PME) ge nerally provides relevant leadership training and education to enlisted personnel, but falls short of meeting the expectations of many officers.

The purpose of this thesis is to examine career progression for ground combat arms officers in the Marine Corps Reserve, and to identify gaps between current and optimal career progression. Recent policy changes provide the catalyst for this thesis. On 4 December 2006, the Marine Corps announced the implementation of the Officer Candidate Course-Reserve. At the time, active component manpower practices and historically high retention rates resulted in reduced numbers of officers leaving active duty following their initial service obligation. Those officers who transitioned into the Reserve Component did so at grades more senior than required to fill junior officer billets. These factors combined to create a gap between RC junior company grade leadership requirements and the inventory of junior company grade officers. As the Marine Corps begins to staff reserve ground combat arms and engineer platoon and company commanders at 100% of manning, the requirement exists to develop a career path that provides the breadth of experience and expertise desired in its future leaders. However, challenges arise regarding the early to mid-stages of career progression due to training opportunities and PME requirements being less readily available to reserve officers than to their active component counterparts.
